Milestone #1: In vitro validation of clinical candidate LEAD-452 and specific tumour localisation
Studies have shown that following intravenous administration, LEAD-452 demonstrates exceptional localisation in tumour tissue. Unlike competing IgG antibodies, ex vivo imaging confirmed that LEAD-452 localises exclusively in tumours, with no accumulation in vital organs such as the liver, spleen, kidneys, heart, or lungs.
Milestone #2: Preclinical efficacy with complete tumour remission
Preclinical trials with LEAD-452 showed complete tumour regression in 100% of mice bearing a colorectal adenocarcinoma xenograft. A high level of CD8+ T cell infiltration was also observed in the treated tumours, reinforcing its immunomodulatory potential.
Milestone #3: Specific memory immune response
Animals previously treated with LEAD-452 demonstrated resistance to re-challenge with colorectal tumour cells. This finding confirms LEAD-452’s capacity to induce a protective and long-lasting memory immune response.
Milestone #4: Scalable production and manufacturing process under GMP-like conditions
Using standard procedures, LEAD-452 was produced in CHO cells under GMP-like conditions, achieving yields of up to 2 g/L.
